Using computers as a vicarious mode of living is nothing new to us, however that one would do this says something about a person's moral character - that if there were some way to live life in a simulation then they probably would. Now the question is, is there, and if so how?

Take pic related for example. This is a game where people can actually get married to each other, presumably because they are too repulsive in real life. In this game, there is scamming, hacking, botting, people who pretend to be girls to get stuff from guys, "kill-stealing", and in certain places one pretty much has to pay money to win. This game is perhaps analogous to the career aspect of life.

Could it be that there is a real life analogue for things we see in this and other games?

I like books that explain math in a conversational way. I've read Calculus Made Easy by Silvanus Thompson and How to Think About Statistics by John Philips, and I like the cozy lecture-like feel that these offer as opposed to textbooks.

Anyone know others to help me learn? I'm specifically looking for Linear Algebra now but if you have recs for other math/STEM fields it would still be nice. Thanks /sci/.

Is rad (absorbed energy in a human) linearly proportional to energy of the radiation?
In other words, if I know that 0 Joules from a source elicits 0 rad in a human and 450 Joules makes 600 rad, could I assume that 75 Joules makes 600 rad?

Hey /sci/, I am working on a project and would like to discuss and brainstorm a bit about certain issues and what approach is best.

This is not a "muh homework" or anything like that, just a discussion about biochemistry.

So, say that one wishes to extract an RNA-binding protein out of organ tissue, with the scope of sequencing the bound RNAs, how would one do that?

Let's say that is is for research, not clinical, purposes. So genetic manipulation is allowed.

If it were a cell culture one could create a tagged-protein via recombination and then simply use an affinity column. However is this also possible in organisms like say Drosophila or Chaenorhabditis or a similarly small model organism without using too advanced methods like CRISPR/Cas9?

Using in-vitro binding (produce the tagged protein separately, then just add it to a tissue lysate) is not a good idea since it does not allow to study the physiological conditions.

Any ideas?

I did something similar to that for my senior thesis in undergrad. (Note that this was 7 or 8 years ago by now, so my memory of that project had faded a bit.) I think you're on the right path, but I doubt you'd be using an affinity column to extract protein in this system. If you're over-expressing in a eukaryotic system, say HEK cells, you'd probably do an IP. From there, you could do a footprinting assay with RNAse to identify the RNA sequence bound to your protein.

The scale that you'd be expressing protein in eukaryotic cells on would probably preclude any sort of classic purification scheme, as would be done from recombinant protein over-expressed in liters of E. Coli or insect cells.

I agree with this. If you take recombinantly over-expressed protein and incubate with tissue lysate, you'll run the risk of non-specific interactions with RNA or DNA or other proteins, plus the hazard of introducing any number of proteases to purified protein.

It should happen on 11-12 August and 13-14 August. Too bad I'm working night shift on both days...

Has anyone seen will there be a live broadcast somewhere online? I would really like to see that. I know it won't be the same as the real thing, but it should do the trick...

You might be falling for the "best meteor shower in human history" hype. I have no idea why anyone created that meme. It's guaranteed to disappoint.

The Perseids typically show about 60-90 per hour, and is one of a couple dozen showers over the course of the year. They happen to be fairly reliable in terms of a show, and it's in summer when it's comfortable to be outside. Sp they're popular.

No meteor shower is worth watching on a TV.
My favorites are the Capricornids, in December. Alas, where I live it's usually cloudy and I miss them.

Want to create a potential energy machine for the use of a personal car. My idea in short is at least a two coiled spring system working in tandem to drive a chassis forward. I'm thinking a small solar generator could be included into the design to slowly contract one spring while the other is busy running the chassis' forward propulsion. (I realize a 2-coil system may not be enough) anyways thoughts?

Me here. If I understood you properly it would be very difficult to use a solar panel to generate enough linear force (or rotational depending on what spring you had in mind) to compress a spring capable of moving the weight of a human. I could see a set up with a rack and pinion gear, with the pinion connected to a reducer, and the reducer connected to the wheels. It could work, but you would need either very heavy, or very long springs. If you are trying to power it by solar energy you would have to convert that energy into linear kinetic energy which could, and should, just be used to push your rack and pinion. You would have trouble extracting enough energy using solar, then further more, you are converting that energy to stored spring tension which results in more losses due to conversion efficiency, making the springs not only unecessary, but detrimental to your cars ability to do anything useful.
