The humor of OPM is not reliant on one joke. Anytime he kills something its not even meant to be all that funny. The humor lies in the fact that a man beyond the traditional cultural programming, a man who lives for his hobby, has attained the highest degree of strength. The humor lies in the fact that all the while there is this grandiose unfolding narrative and cast of characters in competition with each other, there remains saitama as the conclusive force always, the guy who couldn't really give a damn. You'd be dense not to see the metaphorical implications in that as well.
Saitama is a narrative tool more than he is a main character. You'd think that every other character and plot would become superfluous but the reason it doesn't is because saitama lacks the desire to make himself well known or understood, he's just enjoying his hobby. This is why its interesting.

OPM is not a parody of battle shounen. Good battle shounen don't treat enemies as the average "monster of the week" but rather as a psychological barrier that you need to adjust to in order to overcome it, and it leads to character development.
