Medusa's easier to get additional NP levels on and I think her attack growth is higher (like if you grailed her to 80 she'd have more attack). Marie is designed more for durability than damage (8200 attack at level 80 which is something like bottom 5 for all the 4*).

Mashu has a personality, and is also sex. According to both Takeuchi and nasu, she's the real protagonist basically. You're just a faceless ghost that tags along, while Mashu is the one who starts out blank and undergos character development until she becomes a real human bean, and completes her character arc at the end.

She gets a lot better during Camelot when Nasu is writing her again, and her backstory is talked about.

Their abilities and specs are adjusted based on the container; relative to their capabilities in life, they might be stronger or weaker, or have abilities they didn't possess in life due to Noble Phantasms, class skills, or things like Innocent Monster. If you want an explicit case of the Servant being stronger, just look at Stheno or Euryale.
