Despite the memes about her NP damage, it's still one of the best options for taking out big Assassin enemies like the Colossal Ghosts or the Soul Eaters. The demerit afterward isn't too devastating either, if her third skill either fails to proc or is still on cooldown.

Still really good for attrition battles. Revelation gives you a bit more damage and NP gen on stall teams, and having a skill that stuns Servants is really nice for boss battles. And there are plenty of ways to circumvent her stun if it REALLY bothers you, especially on stall teams.

Remember when everyone said Altera was bad? Iskandar is in a similar spot right now. His fundamental gameplay design is good, he's just held back a bit by his lack of NP Interlude. For now, he still does serviceable damage and he still helps improve the rest of the NP chain more than the average Servant with his Def debuff and his Charisma+Military Tactics combo. And the stargen on his NP is pretty good for it being Buster.

Nightingale is, in all honesty, not bad
There are advantages on running a berserker support, namely that they have really low crit weight (and her quick generates a surprising amount of stars mind you) so they won't be stealing stars and her skillset is pretty damn powerful on it's own.
Her flaws are obvious, her NP is the definition of niche, being in certain situations exactly what you need and in most just a heal. But she also does things no other support caster can do, Berserker class damage with a pretty good anti-human buff that makes her sting that much more. Her stats are garbage though, she is a glorified 4*.

You should only level him if you like him over Herc if you have him, the one thing he is better at doing is trying to one turn stuff because he can do more damage for one turn, but otherwise he gets overshadowed hard on everything else.
He is my only single target berserker (except vlad, but I don't have good art enablers for him to burst which was the point, he really suffers from not having any real offensive buffs) but I do like using him when there's a chance to and makes me glad I put the time on him.
