You're only able to kill final strike bosses in their respective rooms and at certain stages. You can never kill one of the shield brothers before they both come out, and Omnigul can be killed multiple times.

You get them by killing/picking up light orbs produced by someone who has them.
You can only get infected once, in a single match.
Once a character is infected, they cannot be re-infected.
If you get only one in a match, then that's it.
If you get all five in the match, then, yeah.
