Wait I think I've deciphered it. IM and Widow's 8 hour missions are guaranteed to drop 3 starstones 100%. IM and Enchantress' 2 hour mission guarantees 1 starstone drop, with a 10% chance to get a second starstone, and a 10% chance to get a third starstone. So if I did my math right, a 1% chance the 2 hour missions will get you 3 starstones.

And, Widow and Enchantress' 4 hour mission guarantees 1 starstone, and a 60% chance to get a second starstone, then a 40% chance from there to get a third starstone. So a 24% chance to get 3 starstones from the 4 hour missions?

Does anyone else not have the option to choose who to send in the portal? Every time I hit 'go' it forces me to send Tony first, Black Widow second. If Tony's busy then I just don't get to second BW or Enchantress, and I don't get to send Enchantress at all. Am I missing something?

As long as the portal is active the option should appear near the top of their task list.

Yes.

Think of it this way:

You can do two missions at a time, so in an 8-hour period you could do:
>2 8 hour missions for 6 stones
>4 4 hour missions for 4+ stones (max 12)
>8 2 hour missions for 8+ stones (max 24)

No, the chance you'll get more than one is just small. But even if you never get more than 1 per mission, the 2h ones are still the best option assuming you are willing to get in the game every 2 hours.
