Well, if I had to pick a character that originated in video games, I'd say Keith David's voice for the Arbiter is really something else.
The Prophet of Truth also has an incredible voice in Halo 2. He was voiced by Michael Wincott in halo 2, and comparatively disappointing voicework was done for the character by Terence Stamp in Halo 3.
