To "beat" Ken, you have to consider the following:

- Ken can literally destroy anything , and I mean ANYTHING, by touching pressure points. He has defeated Saitama, even though Saitama sent him flying into space, by hitting his pressure points before that happened

- the only way the above can be negated, is if the opponent has a mastery of pressure points. A fellow user of Hokuto shinken, like Raho, Tokia and Jagi, or the Hokuto Ryuken users like Kai-Oh and Hyo, are eligible. It ahs been theorize that first and second generation Jojos (Jonathan and Joseph Joestar) could use their Hamon to negate the pressure point damage Kenshiro can deal, but even hamon cannot stop the brute force of an actual punch from Kenshiro

- theoretically, a near abstract being like Cthulhu, the number π and gravity could take on something like Kenshiro, but since kenshiro's fists operate outside of logic, this is questionable

First of all HSK martial art relies on the "dragon breathing technique", while perhaps not on par with Hamon(Sun breathing technique) I imagine it could still do a lot of damage to DIO if a hit landed. Both rely on similar principles therefore damage could still be done.

The real questions are

If DIO can overcome these two abilities he has won.

I also want to point out despite being a vampire, DIO still has human pressure points and Kenshiro would know the right points to counter regeneration, thus it could work on DIO as well. Removing any form of regeneration from DIO.

How to get into Hokuto no Ken:

Anime for first time, there is a bit of filler but only at the start and half of it is actually much needed character development that manga didn't have at the beginning since it was written as a one-shot due to not expecting success.
It also rotates some early events chronology to make better sense than in the manga.
And most importantly while artwork obviously isn't anywhere near as good as manga, the voice acting, music and direction are absolutely top of the line 10/10 material.

You should also read the interquel set between events of HnK1 and HnK2 that Hara made for 30th anniversary of the series, naturally it's canon.
