Yes and no. The XP you get from the story certainly helps move along the masteries, and every step of the story usually gives you a mastery point to spend or save up. At most you need gliding and exalted acceptance (whatever it's called) from the second map's mastery tree to progress the story. Everything else is there to help you complete the maps and make playing much easier for you.

Rangers with the Druid elite spec are the best healers by a long, long way. Other classes have dedicated healing specs, but none come close.

For tanking, it's Mesmer or nothing.

They looked at what people complained about, that being a strict holy trinity, and then removed it with no attempt to change how content is played or any systems to replace it. It turned everything into one big clusterfuck zerg where everyone is a DPS class with sustain. Truly horrible.

Elite specs changed things up a lot for most classes, though Guardian's isn't particularly interesting. They get a longbow and traps, but everybody still uses greatsword.

Condition builds got buffed to the point where they pretty consistently outperform power builds.
