I actually think he could have been a great actor if he really wanted to be, he was legitimately good in Taxi Driver and it's a shame he never tried it again.

He probably decided that if he was going to be a bit character in his movie, he ought to be a bit comedic character. The whole scene is very funny.

The actor for the part actually didn't turn up that day, so Scorsese filled in for him. There is a great interview about the experience with Martin himself where he gives an insight into just how good Rob De Niro was in his prime at playing off his co-stars- can't find the link tho
