If you want to discuss something that will make everybody hate each other, talk about a political issue that is pointless and largely irrelevant to most state policies, but that people are passionate about regardless, like abortion. This will only have a positive result if everybody in the car agrees, then you can enjoy a Plebbit style circlejerk.

Get together a little bit before a long road trip and come up with an awesome playlist. Do this while drinking. Think up a theme tied around stuff you would see along the way or the ultimate destination. Have someone write down all the songs thrown out while you get hosed and then turn it into a bit of sequence that may work. Then as soon as you guys get in, hit play and rock out.
