The best and "most important" museum in Florida is the Kennedy Space Center Visitors Complex. It's one hour east of Orlando.
A ton of smaller amusement parks that are actually a ton of fun
Universal CityWalk is free
International drive has a ton of tourist stuff
Lake Eola is a beautiful park with a 360 view of the downtown skyline
Also there's a college or two so check out the bars in those areas

Seaworld has some very good roller coasters now. Spent a day at gatorland recently and we had lots of fun. Hour or so from Tampa, which has Busch Garden and some of the Gulf Coast Beaches near there are often rated as some of the best in the world. Depending on what you like to do, the corridor between Daytona Beach, Orlando, and Tampa has almost anything you'd want.
