For New York and Boston you could drive, or fly into New York and take a bus to Boston.
The other route would be longer to drive, and probably cost more by the time it was said and done than a plane ticket. But it's a beautiful drive with lots of unique places to see and I think you'd be missing a lot of great memories.

The Arizona-Vegas-SF-Portland is a superb drive with lots of great scenery, national and state parks. While Vegas and SF are also interesting. It is a fairly long drive with lots to see.
Washington DC is interesting with both the sights and the Smithsonian. The drive up to NY is not very far (well less than a day) pretty well built up all the way and few of the cities have any great interest. So a bit short to fly but of limited attraction to drive.

I really enjoyed Lake Tahoe, it was nice and fresh. I went when it was cold and it was really nice. Not sure if it's worth the detour but if you pass by definitely dip in.

I drove from Vancouver to San Fran via the coast. It was very beautiful, and I think it was well worth it. The Norther part of California with the Redwoods was awesome.
The trip took me three days in total.

This itinerary is implicitly time-constrained since the lines are forces and the destinations don't flow.
The worst part about Niagara Falls is that there is any human object within a 1hr walk tarnishes the falls. The purpose-built tourist facilities (hotels, casinos, burger buffets, parking structures) are lowbrow enough to make you regret the stop. Get a jug of water, a loaf of bread and drive directly to the public park. Actively seek a rustic experience or avoid the site entirely. The natural beauty is glorious but the towns on both the Canadian and American sides are vile and a destination "X" is questionable.
