So I had an idea for an encounter wherein the players are split up, and through an illusion end up meeting back up but they each see each other as monsters and fight each other. I'll overrule any player deaths so that no one actual dies and chalk it up to it being an illusion. My question is, how should I handle the combat so that the players don't immediately realize they're fighting each other without sending each player to a separate room?

If you can't trust your players to do it straight and not metagame, the problem is the players, not the situation.
Oh, that's not the problem at all, I was actually hoping to create a plot twist for the players as opposed to just their characters.

You are going to have to employ intricacies, and have everyone play their game separately.
You could fairly easily have everyone enter an area and see hobgoblins, and mediate in, but you will have to employ some 4th dimensional warp fuckery on your gadgets to do it.
