What are the must-haves, what would you like to see in a comfy setting, and lastly, how can I have entertaining adventures in a comfy nonlethal setting?

A comfy setting that evokes a wholesome, warm feeling, like a fireplace on a cold winter night, a soft matress and warm blankets after a hard day's work, or a group of happy children playing hide and seek in the woods.

Literal milk runs.
Hazards include friendly doggos and other animal friends slowing you down so you miss the ice cream man on your way back to base.

Basically Pokemon style. Nobody actually dies, not even the bad guys/monsters. Either once their HP is gone they retreat, or they faint.

Also, replace most of the combat with something else. Like puzzle solving. So when your party gets to the treasure, instead of fighting a final boss for the treasure, the party has to take the four different colored vases sitting in the center of the room and place each one not only in the proper alignment with North/South/East/West, but also in proper alignment with Fire/Water/Earth/Air.

Only then will the magic around the treasure chest dissipate and allow access inside, without simply teleporting itself or the party away.

OP believe it or not I've been thinking about this a lot. Here are some comfy rules:
1. Some of, or all of the PCs have longstanding connections to their family, or people from their childhood.
2. The game takes place in a small setting. It should still be interesting and diverse, but it makes it cozier to know that these characters are exploring a really diverse, interesting small town or nation or something, than it is if they're on some world-hopping journey.
3. Subvert very serious Tabletop Gaming tropes to make them more down to earth. If some Kobolds have had a sacred dragon stolen, give it a name like "Spot" and show the players how its handler has a pet-like affection for it.

Comfy is just the perfect balance between a benevolent and believable world. The conflict can be lethal and high-stakes, but ultimately the players should believe that all problems can be solved with enough effort and that the world works fairly. It should be a place they want to see themselves in, and that will ultimately encourage them to work better with their characters so they can experience it vicariously.

That being said, assuming Fantasy, they're are some ways you could do it...
1. Not Japan Dojo crushing
2. By the waters of Babylon traveling...or just you know Ryutama.
3. Lowryn without the murder elves, instead you get baker/ toy making elves...

Personally I wouldn't mind Monster Musume style setting where you're govt. Agents trying to reintroduce Fantasy creatures into the world...but I'm afraid someone would to try stick they're dick in something...

As others have mentioned, slice of life with low-level conflicts can be comfy. Think of consequences other than damage/death.

>You are all students as a wizarding school, the game is about daily life in the school. Gameplay is about the power of friendship and learning.

>You are chefs at the royal palace. You must find and source rare ingredients, come up with recipes, and put on banquets for the king. (This could easily be a session of Maid)

>You run an alchemy shop together. Gameplay is about earning profits, securing exclusive contracts, exploring the woods for potent herbs, etc.

The nice thing about these very local, low-level games is that it's much easier for you as a DM to populate a small area with good NPCs, interesting locations, etc. It's much easier to make a small town feel "alive"

Maid rpg was very comfy for my group.

We've also played pathfinder with just a lot of nobility convincing and sneaking around noble's houses doing more political stuff than dungeoneering and traveling, which was pretty comfy. The whole campaign took place inside of one city, and we had sessions that were parties and ballrooms and stuff. Our actual speaking and roleplaying was a big part of our success or failure, rather than the dice. It was very comfy.

Slice of life adventures, hijinks at royal parties, inept witches. You can even still have a villain, just make him a rising threat to the world or something that needs to be stopped before he becomes powerful enough or motivated enough to start wrecking scrubs. make him redeemable though, that's always comfy.
