So I got my players to agree to a generic fantasy setting they know they must pass a great test to assend to the sky to fight with the gods. I'm gonna make em space marrines. Any tips, ideas, books? Also general 40k campaign stuff.

Nah, you are set. Using Warhammer Fantasy Roleplay 2e as your base will be fine since the stats translate between systems really well.
Same with using the Deathwatch book and just say "I'm using the system because it allows you to be big badasses."

Don't allow them to have any mutations, they will have to be all male human characters and you'll need to try and keep all of the PC's young at chargen .
All of that is just so that they'll have a chance to survive their selection and initiation.
Female human or any gender of Abhuman may get to become chapter serfs, but that'll depend on the chapter. Xenos and mutants would be purged.
"Magic" user classes can be considered as unsanctioned psykers- those who have little to no corruption and a strong will can be inducted into the Librarium, otherwise they get sent on the Black Ships...

I get the lore constriction here, but if I started playing a fantasy game and the GM said "only male human muggles," then I would be pretty disappointed and probably wouldn't be interested.

If OP had said that the PCs would be getting inducted into an Inquisitorial warband, then anything would go. The best way to get a fit here would seem to be for the characters that aren't potential marines to be given a "crash course" in Imperial lore, then redesigned as (DH1 ascension exp level) Throne Agents, to give them a chance to keep up with the marines.

Also,Space Wolves would probably work the best, since they are already drawing from a feudal world. I read a online scenario a long time ago that had OP's gimmick, the players would fight Wulfen mutants and the geneseed was in a chalice inside an ice castle. When everyone had drank from it, the gm was to just roll some dice, say "oh. damn." And then end the session, without explaining anything.
