What factions might there be in a historical, prohibition era game? The mafia? The bootleggers? The government? Who do the players work for?

Crime syndicates, at least in the states, tend to revolve around heritage.
Italians are the ones you usually hear about but I think Poles and Irish were known for
it as well, I think there were possibly russians too.
If you wanted to go west coast you would have Chinese, Mexicans and Blacks, I don't know if there were any big time Japanese or Korean gangs in U.S.
Besides the gangs the other big factions would be Law based ones. Local Police Force,
CIA, and FBI.
Which ones you pick for the players to be part of is completely up to you.

Don't forget Pinkertons and the like - private eye agencies got hired for a lot of things pre-WW2, from catching counterfeiters to chasing rum-runners to breaking up unions.
