As everyone else is saying, obviously don't flirt during the game. Go over, GM a great game, and stay afterwards (slightly later than all the other guests) to help her clean up the food and gaming materials. (If the others are staying just as late, accidentally leave your keys on your chair before you go and come back for them a few minutes later.) If there's flirting to be done, that's the time to do it.

Started a game for this couple, and the second week three more players came. I am suddenly DMing for three cute girls who have never played RPG's before. Jackpot, right?
Here's the deal; no flirting during game time. Especially if the players are new. Focus on making a great game, being impartial, and giving the best possible first impression for these people.
If this girl and you really have something, you'll surely talk a little more after the game, right? And by being a cool, professional DM who provides an awesome game, you gain at least a little value as a person, seeming creative, as socially adept as a D&D nerd can be, fair minded in adjudication and fast on your feet. If you pull that off, it can't hurt.
There's no guarantee you and she will get together. But even if not, as long as you are delicate, you may win a good group.
