What is the best system for settings similar to Bloodborne?
I haven't played Bloodborne and I know next to nothing about it, but the answer is Lamentations of the Flame Princess.

Well there's soulhunters, it designed to do souls games in pnp form, it's rules lite, don't know whether you consider that a good thing or not.
Personally I think D&D 4e has enough of an emphasis on tactics and positioning to work well for any of the souls games. That's only if you want the players to be of a similar caliber to the hunters in bloodborne though, not if you want the game to take place in a different space in the same setting or otherwise be about different things, what did you have in mind OP?
