I think one of the biggest hurdles with augmentation is maintaining a homeostatic baseline of biochemical operation after any alteration is extraordinarily complex. Look at all the work governments have done attempting to create "super soldiers" or the creation of those nootropic "genius drugs" like racetams and such.
