There are diseases like Klinefelter's syndrome where the chromosomes don't separate properly during meiosis and you get people with an extra X or an extra Y chromosome. You can end up with
X (Turner syndrome - short woman)
XXY (Klinefelter's - androgynous man)
XYY (normal man, but slightly increased height)
and so on. As long as you get one X chromosome, you have all you need to be born alive.

If there is no science behind transsexuality, then it is by definition not a (mental) disorder.
Disorders are caused by phenomena that affect our bodies, some of which we can't explain fully yet. But if there is no science behind it, i.e. you cannot identify something that causes them to want to be in a different body, then it can't be classified as a disease.
The word you're looking for is belief. Like people who believe in homeopathy don't have a disease, they just believe it works.
