Optimistic response? In the next ten years maybe perhaps.
Realistic response? Not until shit down here gets so bad that we HAVE to. There's not much point to sending fragile meat sacks to alien worlds when we can send cheap, expendable robots that can gather more-or-less the same amount of data.

You know it's kinda funny listening to spaceX and Mars one telling us they are gonna land in Mars in a few years, like 4 or 5 years from now, and that they ate gonna stablish a colony, and people how really believe them. Mars one is a joke, a social experiment. They say they are gonna hire all the rockets and stuff for living in mars to private companies (who? There is no such technology like that yet). SpaceX is more serious and Elon Musk is crazy enough to even send himself to Mars but SpaceX technology is still far to do it. So prepare for listening excuses from Mars one and SpaceX for not acomplishing what they said. Nasa is working on it but they would need more social support(more money). Kinda like in the 60s and apolo missions but for that we'd need another cold war and space race... we'll see.

In seriousness, probably in the 2030s or 40s. By then NASA will have a moon station. It's a very slow and methodical process, a lot of other things (SLS, Orion, Altair, moonbase, interplanetary stages, orbital fuel depots, martian lander) have to be designed, built and tested.

NASA has money, it's just that only recently (specifically, due to the STS retirement) have they been able to begin the process of processing all the red tape that goes into making a new rocket. They're on track now, but previous decisions in the 00s pushed back space exploration by about a decade. Shit happens.

And generally, I would be very surprised if it happened before 2030. And even then, it won't be a "colony" but short visit missions with high death risk.

We're pretty much locked on the path of wasting Earth's resources for consumerism, e.g. momentary material distractions. We'll burn through all the easily exploitable resources and will end up having nothing to show for it except for throngs of people with urges and wants that are getting harder and harder to satisfy, regarding the supply side, and a correspondingly diminishing willingness to allocate means toward other goals as resources dry up. Knowledge for its own sake isn't even in the top ten of our priorities. If space exploration still hadn't that tinge of national prestige, we wouldn't be doing it at all.

The dream of interplanetary manned exploration or even colonization has been stillborn from the start.
