Practice.

First, learn how to sport fight - Boxing and some sort of ground defense.

Then take a security/bouncer course, which teaches you how to deal with street violence.

Take a course in more general self defense - those Krav Maga schools have this.

Get conditioned.

Go to bars and pick fights with really weak, small and very drunk guys at first to build up your abilties.

Learning what moves you can and cant cancel out of is pretty good. Learning to use your super bar when needed and is good. You should also try the trial mode because it does help you get the ropes of it.
