I know a guy who works with CRISPR-CAS9 and im intrigued. I plan to go back to school and study this shit myself and become a god among men.

Gene editing doesn't make our tissue and DNA any more resilient. It still wears down, it still needs to repair itself, shortening telomeres each time.

If one could produce mutations that confer maximum advantage with minimum disadvantage. Having massive muscles wastes energy and puts strain on your skeletal. It gives you more strength but you burn fuel at a much higher rate like a muscle car. We could probably do a lot of minmaxing though, improving the efficiency of various systems like oxygen uptake, telomere regeneration, retention of information. Nature after all is not a perfectionist, you just evolve with the bare minimum traits necessary to retain a stable gene pool and nothing more. I'm not like a PHD or anything, just started working towards a BA in Eco/Evo this year, still essentially a layman. As to organ rejection it's possible I suppose but I think the solution that will become mainstream in the next decade is collagen scaffolding combined with organ cloning. The principle is that you take cells from the original organ and then 3D print a model of it in collagen, the tissue which forms the underlying structure of organ systems, then grow the cloned cells over the scaffold. The result is a clone organ which is made with the person's original DNA, this will avoid pretty much all cases of rejection. If the person is suffering from a genetic disease they could also presumably receive a genotherapy tailored to overwrite their naturally damaged DNA.

The organ transplanting thing I'd bet will be common in a decade or two, genotherapy really depends on whether or not other superpowers start allowing the research/use of it, if so we'll jump at it, if not I'd suspect that people will spin their wheels endlessly in moral and ethical questions for quite some time.

The organ transplanting in particular I was interested in primarily as a method to extending human life. After all, what are we if not meat machines? As to extended longevity, the brain is a real pain in the ass roadblock, specifically the bundles of nerves deep in there responsible for long term memory. Unlike other nerve tissue they don't refresh or regenerate once their patterns are established (if they did you wouldn't have long term memory). The issue is that those nerves would have to be replaced eventually or you'll run out of them even if every other part of your body and the rest of your brain tissue is artificially replenished. This leads to a bit of an issue which can only be combatted in two ways to me. One is human/computer interface, you download your 100-ish years of memory onto a robust hard drive and then regenerate your long term memory nerves, retaining only the patterns which result in your personality, or you do away with the human form and move to one with much greater long term memory capacity.

No idea about synthetic neurons or optimized neurons, but I do think that human/computer integration is an inevitability. We already do it right now, sure the computer isn't connected to our bodies but it does act as an extension of our memory and learning aid. Right now, crispr is only useful for adults with diseases associated with certain immune disorders that can be cured with allogenic bone marrow transplants. For example you edit BM stem cells ex-vivo and you ablate the patient's marrow and transplant the edited cells.
Targeting other stem cells are much harder. All you can do is inject your vehicle to the site and hope for the best. Very unreliable and I don't see much future in it right now. China, however, could genetically engineer super humans by editing babies.

Also segments of "functional" DNA do not break off.. it's just the telomere (TTAGGG) sequences that break off... even if the cell stop reproducing it still has a lot of TTAGGG repeats - just less than it used to and that somehow changes the whole functioning of the cell (how exactly this mechanism works is a bit of an unknown)

CRISPR as a gene editing tool is great for researchers who just want to muck around with model organisms (E. coli, arabidopsis, zebrafish, mice, etc.), as it is cost+time effective and it doesn't matter if it goes off target as long as you get one or two successful transformations, but it's just too much of a risk to use in humans unless it's a life or death situation.

The world needs distinct classes. We always need the dregs, the trash that take on jobs like store employees and waiters. We always need people to work in sewers and collecting trash. We need these people who are incapable of getting a better job to settle for menial labor. Can you imagine how quickly cities would literally stop functioning if garbage collectors stopped working because they are suddenly able to become super athletes or rocket engineers?

If we were to ever begin gene engineering it would have to be extremely selective. Largely restricted to the upper class and soldiers.
