You should never convert lossy files to loseless. You'll get noticeably reduced quality. Always, always do loseless -> lossy.

mp3 is lossy. If you have an mp3, you have already permanently lost quality from the original recording. you can buy flacs from hd tracks, download them from wherever, or you can create flacs when you burn from a cd.
You can use Exact Audio Copy for cd audio, dvd audio extractor for dvd-audio, and makeMKV for bluray audio
