Princess Luna was sent back to the moon right after Elements of Harmony so Celestia could still reign supreme. The NuLuna is simply Nightmare Moon's essence extracted from Luna and reformed to be good.

The Royal Sisters and Discord are all celibate, due to being 1000+ years old.
It's not that they've lost the ability, it's that with all their wisdom and all the things they've seen in their life, sex just seems trivial to them, and they feel a disassociation to it. There are other individual factors.

Celly in particular felt obligated to be celibate. Discord and Luna were both absent for a millennia, leaving her the only immortal on the planet. Ponies looked up to her, trusted her, saw her as a white goddess and symbol for all that is good and pure. . . Even if that wasn't ENTIRELY true.
And that, (combined with the fact that she did not maintain close friendships, for fear of losing the ones she loved,) made her decide that it was best to become an asexual entity. Both to set an example and serve as a shining beacon of inspiration, and because she thought it couldn't reasonably work out another way, anyway. Her motherly inclinations and enormous capacity for love caused her many hours of longing and heartbreak, but she never let anyone know it.

Luna was pretty asocial before her banishment, and in fact that was a large part of why she became Nightmare Moon. Back then she was very lonely, and only occasionally had sex with Celestia and a tiny handful of encounters with others she felt for certain she was comfortable with. She constantly yearns for intimacy, yet is extremely selective and dismissive about the subject.
Her natural sensitivity means she needs to feel completely, 100% secure with whoever she has sex with. If she doesn't, she won't do it.

Discord used to be sexual, but he was always aloof about it. He's more detached and more of a natural loner than he appears to be, which is partially why he never had any friends. Another factor is that, while he DOES want friends, he fears any actual intimacy, though he would never tell anyone that.
So he has had sexual encounters in the past, but infrequently. Now he's just old and world-weary, and frankly has other things on his mind at this point in his life. And Ponies aren't as attractive to him as his own species, anyway.
He gives every outward appearance of a stubborn jackass who takes immense pleasure in inconveniencing others. And for the most part, that's exactly what he is, but his character is deeper than that. He has a (very,) hidden compassionate side, and feels very weird and alienated, which is why Fluttershy is so fond of him. She knows it.

The three of them are all at the point where they will most likely break their celibacy, but it'll take a while to warm up to the idea.
