Twilight. It would have the biggest impact. If Celestia or Luna dies, the other takes their place. But if Twilight dies, then AJ, Rainbow, ButterBall, Rarara, they all turn inside out. Starlight loses her mentor, Celestia loses her student and successor, Sunset loses the pony running the barbie portal, everything goes upside down. The system turns on itself. This is the promise of horse land.

Lightning Dust, make an episode how she was kidnapped by an evil sorcerer as a child and given the 'loser' name (in my headcanon the ponies' destinies can be influenced by the names the prophets give them in childhood), spent all her life trying to achieve something grand to make up for it, got into the WB Academy and suicided when she got kicked out of it. I'm not even a LD sympathizer, she was a massive bitch, but that would give her character and make a great episode or two.
