Dash still needs to fully become a Wonderbolt, it would be nice to see where the CMC go now that their goal is to crusade for other ponies cutie marks, and of course Starlight is going to be Twilight's pupil. After that I'm not too sure what the season's twist could be. It went from the letters from Twilight, to ones from the main 6 + CMC and Spike, to that scroll in S3 (I guess), to the box, and last season the map.
With Starlight being a pupil maybe this season will be about them all teaching friendship to other ponies? Be cool if perhaps letters were sent out to Twilight's castle addressed to her and her friends from around Equestria and they had to figure out who to send to solve them and then show that. Though that would kind of defeat the purpose of the map unless they had that going too.

There are some people in the world that know what to do when they see parasprites. That and they don't multiply out of control in nature, so I don't see any reason they would ever manage THAT.
