The best ani/m/e is Tengen Toppa Gurren Lagann. Flawed but its strength help it achieve true greatness while making homages to the finest moments in /m/ history.

Are you asking which has the best story? Which has the best animation quality? Which had the biggest cultural impact/became the most popular? Which one has the fewest flaws and narrative crutches? Which one is the most engaging?

Impact on the mecha genre itself is a meaningless qualifier, because unlike most genres and mediums we can pinpoint the exact moment that mecha anime came into existence, so obviously the very first one takes that cake regardless of any other concerns.
