Noob here, want to read Crying of Lot 49. Is there anything I should know/read before I start?

Not much, it's very accessible for Pynchon. As long as you're aware that Pynchon is an author who is very playful with the prose - plenty of word-play, false clues and leads, abstract metaphors, stream-of-consciousness, etc - then you'll be fine. In fact, The Crying of Lot 49 is probably Pynchon at some of his best while also being at some of his easiest to read. Take it slow, think about what it is he's trying to pull you through and you'll be fine.
