Depends what kind of skirt. If it's a kilt then that's somewhat acceptable. If it's pink, frilly and covered in glitter then you're probably permanently gayer from having worn it.

It really depends on where you are. In my university and the area a round it, skirts are pretty gender neutral.
In some other places (I live in Texas) I'd be fairly likely to at least get stared at, if not accosted and otherwise harassed.
So basically do it where you can be reasonably sure you're safe. (LGBT spaces and college campus for example are a pretty good bet)
