If we're talking about handgun calibers used for self defense, remember that all handgun calibers are bad at killing people, and that it comes down to shot placement, comfortability with round performance (recoil, cycling, weight), capacity, ammo price and availability, and comfort carrying a gun chambered in that caliber. Shot placement goes above all else.

If we're talking about rifle calibers, you must must MUST have the proper barrel length to match the caliber's muzzle velocity as well as a recoil spring that will correctly cycle that ammo's rated load. That said of course the larger the round the greater effect it has against soft tissue and armor. Intermediate cartridges like 5.56 are more likely to tumble around inside their target, which is what's deliberately sought after since they don't produce a larger shockwave inside soft tissue like 7.62x51, which can produce incredible amounts of tissue damage all on it's own. It's more about understanding what different levels of caliber sizes have to do to be effective.
