I'd say it takes a millenium before Mars is habitable and self sufficient. Something like Antarctica is currently ( some isolated research stations) will hapoen before that.

I doubt having a living crew would be feasible. Some fancy technology, like hibernation or perhaps having only cell samples and raising the colonists in tubes at the location would require smaller load.
