In what program? I need it to have a crisp look to it and I will also be printing it out. As long as the photoshop Dpi is 300+ could I make a print out of it that looks extremely crisp?

I'd go for 600 DPI but sure, why not.
You can make it look as crisp as you want, I don't see why not. I have no idea if using a hard spray tool can work since stippling is based on getting the dots closer or further to create values and I assume a spray tool would have the dots overlap. I don't know if it is possible to make a custom brush that keeps the dots separated, but I think it's pretty easy to get there.

just make selections and play with the borders, then go over it again with an inker brush
