Eyes are distorted and moved on every face from every angle differently. mouth too.
Under chin area basicly doesn't exist and is hidden by any means possible.
Anime figures are designed to look good only from right angles, often they do hide the face so you can't see it from wrong angle.

That's more like a sphere than an oval. When you divide the circle into thirds, the center of the circle in the side plane will be the brow ridge, top will be the hair line, bottom will be the bottom of the nose, another third down from there will be the chin, bottom of the eyes will be in the center of the head, and between the chin and the nose will be the bottom lip. If you're distorting the face it'll look more natural to exaggerate the bottom than otherwise.

Modern 3D animation Disney characters don't ever change their skull shape. The facial muscles around the eyebrows, mouth and nose are elastic and done with stylized exaggeration in mind, the cranium is always stationary and never changes form. That's some old 2D animation Tom & Jerry shit.

it gives more liberty in expression, character customization and movement
if you have the skin separated from the bones, making an elderly or fat character would be just as easy as the default one, with the bonus of each fold of skin acting on their own physics, and not of the bone's

Flip your drawing, it has lots of problems and that should help you to see them. Everything seems deformed and to be falling on the left, which obviously isn't right.
Secondly, use references for your bodies, you'll need it. Might as well do the same for Anime faces because it looks pretty bad.
Third, use thicker lines and try to remove your chicken scratching.

There's a lot of things wrong with this, but it all stems from a lack of foundation.
If you want to improve you're going to have to go back to the basics.
Before doing any of that though, lower your brush size, it's not doing you any favors at this point in time.
It's great for someone who understands anatomy but you're not at that level yet.
All it's doing for you is making your lines more vague and hiding your mistakes from yourself, sure it might look cleaner than when you chicken scratched >>2478434 but it's basically the same thing.
The main reason why your line art isn't clean isn't because you're not used to drawing with a tablet, it's because you lack confidence, and you lack confidence because you don't understand what you're drawing.

First it allows you to know where you stand in your knowledge of anatomy, highlighting where you need to study more.

Filmicly,even on a camera with a very low aperture, the background robot would still be in focus, or at least the leg she's resting on, Because it comes into the plane she's at. SInce it's anime, I suggest just drawing out the entire thing, but do it in small steps, knee first, then leg, finally body, so that you can look at the big picture and see what works.

For example, the elastic part of her socks should be sticking out a tiny bit to make it seem like an actual article of clothing. Nice! You are getting better at making them look actually big. Push the perspective even more tho, usually with drawing giants you want to convey a sense of being overwhelmed and pushing the perspective works wonders for that. The people in fromt should blend in a little more, especially the 2 on the left pop out too much, they are separated from the background with black outlines and the colors are stronger than the colors of your focal point which is the giant loli, that makes them stand out even more. Hey. I've been watching you for a while and I'd like to drop a piece of advice as well.
To add to what anon said, I've noticed a trend in your giant loli compositions that I think is hurting and holding you back, that trend is that the loli is always seen in her full form, she's always the immediate focus and everything around here is "just there". The only reason we know she's big is after looking for a bit and noticing "these things just there" are buildings and such. It's still nice but it could be so much more.
What about thinking outside the box and making a drawing from inside a house for example? Seeing the backs of random people looking outside their window and seeing a giant loli? In a view like this the loli will immediately strike as being big, but more than anything you'll need to refrain yourself from absolutely wanting to draw 95% of her body. In the same vein, (re)watch some Shingeki no Kyojin (or look at the manga), I'm sure it'll give you a couple ideas to spice things up.
My post is a mess I know but whatever. Oh and it's not like I'm saying your art is shit or that you should change everything, nah not at all. I just think you have the potential to do and experiment more things.

As far as the angles goes, I'm pretty satisfied with the picture's angles because these characters have very very little art of them
This means that for a beginner, it is very difficult to imagine completely new angles for the clothing of these characters (the girl holding the rifle, there's only one art of her and it's sort of frontal one)
But besides that, any big anatomic issues to take care of before coloring?
Pic will get some minor adjustments on the tank before coloring too I suppose

That's why you need to learn perspective and anatomy. So you can break down the character into some basic forms and then you can rotate them to any angle you want
