Probably not because it will lack power for spinup. If for some obscure reason the phone gives enough power - sure.
But then if you have NTFS or some macfag FS on your HDD you are fuged if there are no packaged drivers for your OS (Android?).

I've done it. one of my 2.5" enclosures has a 160GB exFAT formatted drive in it. It also has a separate plug for power so when it's used on my PC it needs 2 USB ports. When it's plugged up to my phone I simply use a spare Samsung or LG power adapter and plug it in there and it powers up and works fine.

if phone supports OTG that yes. however, you'll probably want a Y-cable to connect a powersource too, as a USB-hdd often draws more power than OTG provides (it's often capped at 100mA)
