Is there an audible difference when listening to audio via a laptop with a DAC connected via USB (music is accessed from an external HDD), versus using a DAC with a DAP via USB (using the external playback function)?
Maybe I'm not phrasing my question well, but I was wondering if the sound would be worse from my 8 year old laptop because of a possible delay of accessing music from a folder on my external HDD as opposed to my DAP where the music is stored internally AKA faster access.

I dont see how that would worsen or improve sound quality.
If you are worried you could always add a buffer time to your stuff that way even if there is a delay from transfer it shouldnt impact you.
I have a 2ms buffer for DSD 128 myself.
laptops tend to be huge noise generators and if your dac doesn't have properly designed isolation this noise can pass through dac. DAP is much simpler device and probably generate much less noise.

I think I will purchase a mini usb OTG adaptor for my DAP anyway, because my laptop fan makes way too much noise to appreciate my open-type headphones.

No, that shouldn't be an issue, but depending on what DAC you have, if it's an entirely USB powered model and your laptop's USB power is very noisy, that noise could actually end up on the DAC's output too. If your DAC has its own power supply this is much less likely and it's of course impossible if you're not using an electrical connection between laptop and DAC, as in you're using optical TOSLINK.
You most likely won't hear any difference, if there's noise coming from your laptop it will probably be quite obvious. If it's not you shouldn't really worry about it.
