Unless you're replacing your CPU, I wouldn't do that too often. Thermal paste shouldn't need to be applied more than once and after it is, it generally is fine to leave as-is. As long as you didn't get any crud in the paste or anything, you should be fine.

Run some stress testing and monitor temps prime95 abd hwmonitor. But yes, your temps will increase, not sure how much but they will. You should always reapply paste when you disassemble heatsink ftom cpu.
