Not really possible, MP3s strip unnecessary data off of the source material, thus making them smaller but making the quality a bit worse in the process, that's why it's called a lossy format, and I don't think it would be possible to add this data back.

No - mp3 is a lossy format, you lose data when converting from the source to an mp3

converting the mp3 to flac will not regain any of that data

It would sound better than if you had converted the video from mp3 and then to flac. The quality would be exactly the same as the quality in the video due to flac being a lossless format (no data/sound quality is lost when converting to flac).
