There's no easy answer to that question.
But disregarding the multiple different versions of both characters and whatever nuance there might be, it's probably best to say that they're the same guy, but one is insane. Galvatron in the film acted pretty much like a slightly more competent megs, so it's safe to assume the thing that drove him crazy was losing his connection to unicron, which 'cron probably did on purpose. It's a very Unicron thing to do.

In the G1 cartoon Galvatron was an upgraded dying Megatron who eventually went insane.
In most of the comics they are different characters. Either Galvatron is some sort of herald of unicron, or he is an ancient Cybertronian warlord.
Megatron is usually much the same as his G1 cartoon counterpart, only more competent. In the IDW incarnation he's more morally ambiguous. As the comic delves into his backstory, you learn he's actually a sort of revolutionary with a violent backstory. You see him rise from being a lower class miner with a dream of changing an oppressive society, to a ruthless despot who's motto is "peace through tyranny."

Not really. That's only IDW.
In the Marvel comics he was still Megatron-reformatted-into-Galvatron, but he time travelled so he was in the same time as Megatron, so both existed, but still former Megatron.
