That's hard to answer. Old Looney Tune and Merrie Melodies are considered classics. Avatar the Last Airbender might be the best in terms of telling one whole collective story rather then being episodic like most western cartoons.

Ed, Edd n Eddy. Mixes old Looney Tune slapstick with it's own slice of life simple aesthetic and stories, the end result being a lot of humorous, heartwarming and even ingenious moments.

Don't watch much anime but Madako Magika is a great show that's subversion of the "Magical Girl" genre.
