We might see more of Ludo's family history, how/why they became royalty. but I'm not sure if we'll see Ludo's own backstory more than the implications we got. If only because his parents and siblings constantly being dicks to him might be kind of much for a kids' show. Maybe a flashback with Dennis.

Toffee's backstory seems pretty much confirmed in the S3 premiere, at least in relation to Moon. Though we could get more too if/when we learn how he knows Eclipsa, if that's not revealed there.

There's no way she won't be treated as a villain by everyone in the greater setting. The Magic High Commission almost shit themselves just because Star was reading her notes unsupervised.
That said, she is in a unique position where most of the characters we're actually focusing on probably wouldn't treat her as a villain outright, on their own judgement. Star clearly doesn't take her seriously and seems to even admire her a little, Marco is a human who would probably defer to however Star ends up dealing with her (though more cautiously, considering he was actually possessed), Ludo is almost certainly ignorant of her entirely, Buff Frog probably is too but between his competence and his good relationship with Star probably wouldn't go picking a fight with a Butterfly Queen.
Moon isn't prejudiced against monsters and seems aware that the "monster love" thing has demonized Eclipsa to some extent, and her reactions to Star meddling with Eclipsa have been more restrained than the MHC's or River's, though she's still clearly concerned. She would oppose Eclipsa more on the basis of her being a legitimate threat to the Kingdom and Star's wellbeing than whether or not Eclipsa was/is a bad person. Not even getting into the fact that the thing Moon apparently made a pact with for her "Darkest Spell" was probably Eclipsa.
As for Toffee, we just don't know enough about either to do anything but speculate.

In my opinion,
1. Eclipsa definitely isn't all bad. She has way too many sympathy flags, between her clear parallels with Star and running away with her true love. We already have a straight BBEG villain in Toffee.
2. Eclipsa is still pretty bad.
3. The Eclipsa/Toffee connection is a red herring.
4. Eclipsa's role in the story will be a third/fourth party, not aligned to the Mewmans or Toffee's monsters. She will want peace between Mewmans and Monsters, on her own terms, by force if necessary. She will fight both sides while trying to get Star on her own.

Anyone else hoping Season 3 is them building up to a relationship and Season 4 is them dating with there maybe being some kind of required marriage subplot or something?

I feel pretty confident that they'll be "together" before the finale. There's definitely a lot more going on to keep the story/drama moving than romance, so it's okay if they blow that load before the end and take the rest of the time to explore them as a couple.
Even if they're happily together there's still conflict to be examined, without resorting to cheap shit like wacky misunderstandings or cheating. They'd have to come to terms with Star's inevitable future as Queen, and what that would mean for both of them. And there's still the Blood Moon, which definitely isn't going away. We'll probably get an episode explaining what exactly that was about eventually. How would Star feel if she thought that falling in love with her best friend might not be "real", but part of some curse, just another thing in her life that's she's been pushed into by forces beyond her control?

I don't think they'd do this with a serious angle, but I love the idea that's been floated in the last few threads of "Moon and River told everyone Star and Marco are engaged to keep up appearances after the Song Day shitshow."
That has a lot of opportunity for comedy and romance teasing, and would mostly lock them out of doing some cheaper romance drama like "Prince Chad seeks Star's hand in marriage" or "Mermaid thot wants to get in Marco's pants."

You know, there ARE some mysteries surrounding Jackie thay need explaining. She used to live along/in echo creek. That would mean she was, potentially, either homeless (like hair lady) or a mermaid. She has been a presence in Marcos life for years in a non-magical world so her mermaid status would be unlikely. Does her being super poor scan, though? Skater girl whos family we never see and who is almost always in the same clothes despite many others having multiple costumes. That sounds poor to me.

What if the thing "never done in a childrens cartoon" is to depict a character who is living on the edge in a cracked or broken family. No safe, comfortable fantasy for these kids. Jackie lives a raw life and puts on a brave, nonchalant attitude as a defence against the fear she always lives under. Maybe one of her parents is gone or terminally ill and so she suffers with abandonment issues
