Not at all. It's all pretty self contained while mixing lore form the movies, comics, and adding some stuff of its own. There's some shutouts for readers of the comics, but you can go into it blind and be perfectly fine.
Afro Samurai 2 might have been shit, but at least its lazy design made for a convenient one-stop spot to listen to all of Ron Perlman's unique Justice dialogue from the first game:
