This is the reason why Ashi eventually joining up with him will be so pivotal to his character development for the rest of the season. He'll finally form a consistent, stable relationship with another person after 50 years of hell, and one that he he truly sympathize with and relate to thanks to their similar harsh backgrounds. He's finally going to find someone that gives him hope and purpose to live on and continue fighting. That's why there's so many underlying parental themes being implied in the new season (calling the 7 sisters the Daughters of Aku is just one example.)

What if the show's tone follows that of the comics he's referencing? Comics from the 70s and early 80s were light hearted and more cartoonish, and then got darker and darker for nothing more than the sake of being dark and edgy through the 80s and well into the early 90s. But then they started to lighten back up in the 2000s and till today. Comics now can tackle adult issues without being needlessly dark and edgy.

It was indirectly implied in The Princess And The Bounty Hunters that he killed humans; namely Boris, The Gentleman, and Jujunga. I And Am are certainly not human and thus count as either alien or robot, which were fair game in the original series.
However, in Episode XLIV, we can't be 100% certain that Jack actually killed anyone. There was no blood of any type to indicate death and, due to the show's episodic format, we were obviously not going to see those bad guys again.
Many fans of the show have assumed that Jack did kill all of them in the altercation and that it's implied due to CN's stance against Jack killing humans.
As someone that watched the show when it premiered in 2001 and religiously followed every premiere until 2004 (and, obvious, 2017), most fans either had no opinion on XLIV or generally agreed that Jack probably killed them.
HOWEVER, Genndy has been asked about this episode multiple times and - every time - he's shrugged it off, saying that he can't quite remember the whole episode, what his thought process was during production, whether they were actually aliens or humans or robots, whether any died or were just knocked out; basically, he's not giving a straight answer because he doesn't want to explicitly retcon anything and upset any fans who have thought otherwise for over ten years. At least, that's my two cents; of all the things he's been really indecisive about with the new season, it's that.
I think that Genndy just wanted to deal with the impact of Jack killing his first human being on Genndy's own terms. He could not deal with it directly when the show aired on CN because of content restrictions.
I get how some people don't believe that Jack has gone 50+ years in the future without killing - even I, a fan of the show for 16 years, have doubts about that. But I've got faith that Genndy will execute this aspect properly.

Well of course this shit isn't entertaining or fun to see IRL or experience first-hand, but it sure as hell makes for some great entertainment in fiction... especially for those middle-class or wealthy people residing in well-off countries, where they don't really get to experience life's highest and lowest points because of it.

They can't will likely not experience any true hardship in their lives, so they turn towards entertainment to fulfill this. Addressing the 4th wall is a plead to the producers or creators of the TV show. Since the show wasn't being aired Jack/Aku have been in limbo. Trapped in an eternal battle where neither of them can win, because the story didn't provide and ending. Aku is asking for an end to this story, and end to his suffering. (he's so very tired) The story can only end with the hero winning, which from Aku's perspective, means the story won't end until Aku's won. After killing a human being, Jack is no longer innocent. I think it's possible, the story won't end with Jack's success, or more likely will end with a pyrrhic victory.
