Can anyone defeat Karnak in hand to hand combat?

Anyone who can beat Karnak in hand to hand combat would have other weaknesses to exploit. Civil War 2 shows him using psychology as much as his fists.

A LOT of people. People who are simply too fast for him to react to (Spider-man, Quicksilver), People who are too durable for him to hurt (Luke Cage, Venom), People who are simply better fighters (Iron Fist, Shang-Chi)

Keep in mind that Shang Chi's the MU's greatest martial artist but that Iron Fist could still beat him because he has the added edge of magic punches.
There are countless variables at play. The answer is whoever the writer wants to defeat him desu

Anyone who is siginifigently faster or stronger. You might see that a guy has a bad knee, but when you need 2 tons for him to feel it, you're fucked.

He has the Batman effect in that regard. One punch should break his jaw or give him brain damage, but lol your punch has less force if I go at this angle. Then again I just want to see someone use gunkata.
