Actually, Kryptonians originally suffered from a disease that killed them if they left their home planet. There was an alien 'prophet' that had a vision of Krypton blowing up and listened to Jor'El, he convinced a lot of Kryptonians to leave the planet on a arc-ship he brought with him. They all started dying the second they left the atmosphere. There were thousands on the ship and the prophet watched all of them die in front of him and went a bit mad. Superman met him on Warworld.

The reason that Superman didn't die was because he was actually 'born' on earth and never breathed kryptonian air until he was older and his immune system could protect him from it.

It was heavily implied that this was either a bio-weapon used to keep the kryptonians on their world to stop them from conquering the universe OR a means of control used by the elders to keep kryptonians from going out into the universe.
