Do Spider-Man's powers give him the peak physical human condition? Like if he lost them, he'd still be in fantastic shape because of all the time his body had to adapt to the stuff he's done, right?
It's not as if his powers ae a suit of armor or a buffer that absorb every benefit from the physical activity he does. His boddy is a conduit for the power to act, at least that's the way I see it. I think you could say that about almost any physically powered hero if they lost there powers. Maybe not magical, MAYBE. But They all spend there time engaging in repeated strenuous activity that's sure to have most of them in pretty good shape. What you may not have considered is the long term effects of all the injuries they've sustained over the years without their abilities suppressing the pain or repairing the damage. So they might come out of it pretty fucked up all in all but with really great bodies.

It's a shame if it was forgotten because it was a great concept. It was like the next step for Spider-Man, to really master his powers. Not necessarily. The symbiote loves/is obsessed Peter, that's their dynamic, not anyone elses, and I'd be annoyed if they take that unique thing and implant it over to a completely different character.
The reason why the symbiote was so taken by Spider-Man in the first place was becuse he was Peter Parker, a good noble soul that freed it from imprisonment. Gwen is a completely different person, you feel me? So if they give her the same dynamic it'd be as bullshit as giving her black suit the same designs as Spider-Man - there is no reason for it, it's just gifting one character the unique attributes of another for convience and the sake of plot/fan appeal (Silk).
They should really find a different approach for her symbiote, though. Like maybe she doesn't get rid of it like Pete did, or maybe the one from her universe IS bad news and she's justified in wanting to get rid of it. But in any case it is a presumptuous assumption that the relationship between Gwen and her symbiote would be anything like Peter and the Venom symbiote.

The problem with this question is you're asking to rationalize the irrational. Not to be dismissive, because these kinds of things are fun to think about, but at the end of the day we all kind of have to shrug and chalk it up to comic book logic because in the real world there are no super powers, so trying to explain them is eventually going to be futile.
That said, let's play along. The physical powers are easiest to explain. Super strength, durability, reflexes, agility, healing. Increased muscle density/efficiency, bone density, tougher skin covers most of that, with a more robust nervous system accounting for some of the rest (muscle memory/reflexes are mostly nerves.) Maybe metabolism for the healing. The thing with all of those is that they rely on distinct physical structures in the body. That means, there's not really going to be an on/off switch. You don't just wake up one day and lose the majority of your muscle fibers or nerve function, you know? At least not in the way of losing your "powers." So those things would likely stay the same, or maybe slowly degrade over time back to normal human levels.
Then we have wall crawling, which is sometimes a physical trait (hairs) sometimes a metaphysical one (electromagnetism or however they explain it these days.) If it's physical, see above. If it's metaphysical, there might be a way to disrupt the energy field responsible for it.
Spider sense is almost definitely metaphysical in nature, as it's practically (or sometimes literally) precognition. It gives him information that he literally could not physically perceive. So that's something that could be blocked or disabled... Probably.
So yeah, if we're pretending to be scientific, "losing his powers" would probably leave him without his spider sense or wall crawling, but leave him with his superhuman physical traits at least for a while before they degenerated. Possibly he'd keep them though, depending on what mechanism sustains them.

He's have enough muscle memory to be as strong as Daredevil; somewhere around the neighborhood of benching 450 pounds. Probably has good endurance from all those years of intense acrobatics and brawls. His hand-eye coordination is up there as well as his reaction time.
The thing is, Peter needs his powers to operate as Spider-Man. His spider-sense helps him aim his webbing at secure buildings while swinging. And he needs his superhuman durability and healing factor to be able to land on his feet from high speeds. I've fucked up my knee during track and field so imagine the kind of damage a regular man's body would take in Peter's typical feats.
