It's just an anecdote, you don't have to react any way other than you normally would.
I would say you have to just act normal and pretend they didn't say anything out of the ordinary unless the subject actually does turn into being about homophobia in which case you can say something about it. When people talk about their couple, gay or not, I just say uh huh and cool.
