SAI is really good. Photoshop is the industry standard for illustration, but a lot of professionals prefer to work in SAI.
GIMP is watered-down photoshop, but you can make really pretty things with it.
Personally I'd recommend Krita (https://krita.org/) be added to the list - it's a lot like photoshop, but more focused on illustrators' tools. It's not quite as popular as PS or SAI, but IMO it's the best freeware art program out there.
If we're talking paid programs, Clip Studio Paint/Manga Studio deserves a mention. Has a lot of tools for making comics.

So office, holiday, party settings are quite common for these characters. You can do something based of that premise or anything else you might think of.
If you'd like to draw lewd. They're both older futa that are into younger men. Younger or older women/futa are fine too.
If you'd want to draw something simpler or sfw, a pinup works too. You could maybe draw one of them in different clothes (like winter wear for example). You don't need to show that they have a dick if you don't want to
