Isekai is a setting and a trend, but not a genre, so they're not mutually exclusive. Neither series is a parody, either.

Konosuba is a pretty straightforward Isekai that differs by not taking itself seriously at all and by having been written by a very silly author. It's a comedy twist on the formula but not a parody, despite the occasional referential joke. (Similar jokes are present in pretty much any comedy work out there.)

Re:Zero is also just another twist on the standard isekai formula, but done by an author who's a bit edgy. It's not parody either, and I reject the "deconstruction" thing you hear thrown about so often, it's just a different take on the setting.

Konosuba relied on pretty much making fun of shit or some trope or making ridiculous situations or references(last reference I remember it made was Phoenix Wright or Rokka). When people say the comedy is unique or some shit, it's not, even slayers was better. This series is a joke, and not a very funny one after a while. I want my 8 hours back please.
